As such, they lack light chains, are smaller and more stable compared to conventional antibodies, yet possess fully functional antigen-binding capability. - Selection and first pass sequence of ELISA-positive binders
- Small-scale production of up to 6 final positive clones
Antigen Requirements
Antigen Purity
- For recombinant single domain antibody development, >90% antigen purity is recommended for specificity and functionality.
Protein Antigen
- Quantity
- 0.5 mg at concentrations of 1 mg/ml or higher for immunization
- 0.5 - 1 mg for screening (quantities varies based on project please inquire)
- Size Ideally greater than 10 kDa
- Production method Using a properly folded native protein is preferred as sdAbs are adept at recognizing conformational epitopes. Therefore, it is ideal for your target protein to come from an expression system similar to the origin of the protein (e.g. use a mammalian expression system for human or other mammalian proteins).
- Tags It is recommended not to have tags for immunization and large fusion proteins such as GST or MBP should be cleaved to minimize nonspecific immune response. Contact us for your project.
