Methods of Antibody Purification

Several techniques are employed to purify antibodies, each tailored to specific requirements:

1. Protein A or G Purification

Protein A and Protein G are bacterial proteins that bind selectively to mammalian immunoglobulin G (IgG) antibodies. This method isolates total IgG from sources like serum, ascites fluid, or cell culture media. The choice between Protein A and Protein G depends on the species and subclass of the antibody. However, this technique is not recommended for peptide antigens, as antibodies against larger carrier proteins may also be isolated. It is best suited for antiserum produced using protein antigens and for purifying monoclonal antibodies from ascites or cell culture media.

2. Immuno-Affinity Purification

This method isolates antigen-specific antibodies by coupling an antigen to a gel matrix, creating a column that captures the desired antibodies. It is particularly effective for isolating peptide specific or protein antigens and for purifying monoclonal antibodies. 3. IgY Purification

Chicken IgY, functionally equivalent to mammalian IgG, is primarily isolated from egg yolks. The purification process involves removing lipids and precipitating antibodies from the resulting solution. The total IgY can be used directly or further purified using an immuno-affinity column.
